Callus and Suspension Culture Techniques Optimized for Use in Carrot Breeding Studies (Daucus carota ssp. sativus var. atrorubens Alef and D. carota)

In this study, studies were conducted to optimize callus and suspension culture methods for in vitro mutation breeding purple orange carrots. Following this, the developed mutant lines subjected salt stress selection. The study determined optimal agar dose. first germination, 50% root-cotyledon formation genuine leaf carrot seedlings analyzed daily which was continued with 7g/L cotyledon hypocotyl explants from cultivated production mixtures of MS-1, MS-2 MS-3 nutritional medium. second fourth weeks following subculture, percentages weights measured. (1 mg/L 2,4-D + 0.5 kinetin) medium explant found be particularly effective at producing callus. subculture's data on number plants that had successfully regenerated per callus, showed MS 0.2 Thidiazuron (TDZ) best plant regeneration. dispersed calli grown a designed nutrient mixture comprising MS+0.1 kinetin. results obtained optimization steps used ongoing study.
